MiCoCo: Chemoenzymatic synthesis of complex polyketide natural products using biosynthesis enzymes

Abstract:

The application of biosynthesis enzymes in chemoenzymatic synthesis has large potential to improve total synthetic routes to complex natural products.1,2 Particularly in the case of highly functionalised polyketides, typical enzyme properties like high selectivity and mild reaction conditions can mitigate frequently occurring problems.
We explore the enzymology of polyketide biosynthetic pathways with the aim to identify candidate enzymes with synthetically attractive activity. These are
biochemically characterised and studied regarding synthetically relevant features like their substrate tolerance. Synthesis of complex biosynthetic intermediates is also essential.
Progress on the chemoenzymatic synthesis to the unusual reduced polyketide
jerangolid A and its derivatives will be presented.3 We have established efficient routes that yield specific series of biosynthetic precursor derivatives. Several tailoring enzymes and domains from the jerangolid and the related ambruticin biosynthetic pathways have recently been characterised. Progress on their integration into a chemoenzymatic route to jerangolid A will be presented together with the results of in-depth studies of some of these enzymes.3-7
